"All up ons" is a colloquial usage describing popularity. As Strong Bad explained to Strong Sad, "It means they really like you". However, the phrase appears in many places in the Homestar Runner body of work that don't have anything to do with this usage, often where "up on" would be sufficient.
A variation on this phrase has also occurred in the form of "All up ins", which simply means in.
All up ons
