The video game franchise Mega Man has been referenced throughout the Homestar Runner body of work, particularly where 20X6 is concerned.

- 20X6 may be a reference to 20XX, the time period in which the Mega Man games take place.
- Stinkoman's "robot boots" are similar to the ones that Roll, a female character from the Mega Man games, wears.
- Email japanese cartoon — One of the NES endings in the Easter egg is of Mega Man 2.
- Teen Girl Squad Issue 4 (DVD commentary) — The Brothers Chaps compare the Giant Brain Mech to a Mega Man boss.
- Marshie's Malloween Mix-Up — Marshie's death animation is taken directly from Mega Man's. Also, the health bar is similar to the one used in the Mega Man games.
- New Boots — In the Hip Hop Dance music video, the pixellated Coach Z is an edited sprite of Mega Man.
