Popular hip hop trio De La Soul has been mentioned or referenced multiple times in the Homestar Runner body of work.
Matt Chapman listed their album De La Soul Is Dead as among his five favorite records in a 2003 profile.

- Email 50 emails — One of Homestar Runner's commands to delete the email is "de la soul".
- Email monster truck — Homestar's line, "Let's do this like Brutus," is from the first skit on the album De La Soul Is Dead, which itself is quoting the Public Enemy song "Bring the Noise".
- The DVD commentary specifies the reference is to De La Soul.
- Email big white face — Strong Bad refers to the Poopsmith as "doodoo man". This references "Rap de Rap Show" from De La Soul Is Dead, which features a fictional DJ who refers to himself as "the Doo Doo Man".
- Email death metal — One of the words that starts with "D-E" that Brainkrieg sings is "De La Soul".
- Email looking old — Marzipan's line, "Word booty!" is from the De La Soul song "Bitties in the BK Lounge".
- Kick-A-Ball — The page title is based off the De La Soul song "A Roller Skating Jam Named 'Saturdays'".
- Final Hours Playthrough — When a Smell the Daisies action card is played, Strong Bad remarks that it is a daisy age, or so De La Soul tells him. De La Soul's debut album was themed around the concept of the "D.A.I.S.Y. Age" (an acronym standing for "da inner sound, y'all").
