Homestar Runner shows more respect for The King of Town than most of the other characters do, but they do sometimes conflict with each other.

Homestar Runner and the King of Town in Conflict

• The King of Town DVD — When Homestar realises that the King ate his sheep, he embarrasses the government official, while poking him.
• A Decemberween Pageant — The King is upset that Homestar was cast as the King in the play instead of him.
• Bug In Mouth Disease — Homestar eats the King's ice cream, believing it will freeze the bug out of his mouth.
• Halloween Potion-Ma-Jig — The King refuses to give Homestar the powdered herbs and spices, prompting Homestar to snatch them.
• Email more armies — Homestar and the King are the leaders of competing armies.
• Homestar Ruiner — The King of Town is insulted by Homestar showing himself naked.
• Strong Badia the Free — When the King places Strong Bad under house arrest, Homestar joins Marzipan's protest against the King. Also, in Maps and Minions, Homestar tries to stop the King from getting back to his castle and will go after him if he is visible.

Homestar Runner and The Poopsmith seem to have a good relationship, though they do sometimes have bad moments.

• Strong Bad is in Jail Cartoon — Homestar leaves the Poopsmith tied up, and clicks a flashlight on and off in his face, while saying "All right, buster, move it along here! Nothing to see, nothing to see!" and thinking the Poopsmith is Biscuitdoughhandsman.
• Email highschool — Homestar Runner pushes the sweeping end of a broom at the Diapersmith.
• Strong Badia the Free — If Homestar and the Poopsmith meet up, Homestar kicks the Poopsmith in the stomach.

• Email colouring and Career Day — Strong Mad and Homsar are both students in Marzipan's LURN class.
• Strong Badia the Free — If Homsar meets up with Strong Mad in Maps and Minions, Strong Mad punches Homsar all the way to Strong Badia.

Strong Sad and Strong Sad's Relationship

• Email replacement — Strong Bad asks Strong Sad what he would to to "himself" while he is on the couch watching TV. Strong Sad says that he would make "himself" some hummus and give "himself" a foot massage.

• Email highschool — Baby Strong Sad does not want to sit next to the Diapersmith.

Marzipan and Homsar have an interesting relationship. Marzipan seems to enjoy hanging out with Homsar; Homsar himself acts like he doesn't even know Marzipan.
